Kymera Therapeutics KYMR is leveraging targeted protein degradation, a next-generation small-molecule therapeutic modality that harnesses the body's natural cellular recycling system to selectively eliminate disease-causing proteins.
The company is developing lead candidate KT-621, an oral STAT6 degrader, currently being evaluated in parallel phase IIb studies in atopic dermatitis (“AD”) and asthma.
Kymera completed enrollment in the BROADEN2 phase IIb study of KT-621 in moderate-to-severe AD nearly six months ahead of schedule. Top-line data are now expected by year-end 2026, six months earlier than the company’s previous target. Subject to regulatory discussions, phase III studies in atopic dermatitis are planned to begin by mid-2027.
The trial is evaluating three doses of the once-daily oral STAT6 degrader against placebo. Its primary endpoint is the percentage change from baseline in the Eczema Area and Severity Index score at week 16.
In January 2026, Kymera expanded the BROADEN2 study to include adolescents alongside adults. This is a global, randomized, double-blind, placebo-controlled, dose-ranging study evaluating the efficacy, safety and tolerability of three doses of KT-621 in approximately 200 adult and adolescent patients, ages 12 to 75, with moderate to severe AD over 16 weeks. The primary endpoint is the percent change from baseline in Eczema Area and Severity Index score at Week 16. Secondary endpoints will evaluate additional safety, efficacy and quality-of-life measures.
In June 2026, at the Japanese Dermatological Association Annual Meeting, KYMR presented results from the phase I study of KT-621 in healthy Japanese adults. KT-621 demonstrated a favorable pharmacokinetic profile, along with rapid and sustained STAT6 degradation in the blood, with median degradation of at least 98% at both dose levels. The therapy also showed a favorable safety and tolerability profile. These findings were consistent with results previously observed in non-Japanese healthy adults and patients with atopic dermatitis.
Positive data highlight the candidate’s potential as an effective oral alternative to injectable biologics.
KT-621 is also being investigated in the ongoing phase IIb BREADTH trial in moderate-to-severe eosinophilic asthma. Kymera continues to expect top-line data in late 2027 and has initiated an open-label extension that permits eligible participants to receive KT-621 for up to 52 additional weeks.
The candidate has received Fast Track designation from the FDA for the treatment of moderate-to-severe AD and eosinophilic asthma.
The successful development of the candidate will be a significant boost for the company.
Competition for KYMR’s in Protein Degrader Therapies
Arvinas ARVN is a clinical-stage biotechnology company pioneering PROTAC (PROteolysis TArgeting Chimera) protein degradation therapies.
ARVN, in partnership with Pfizer, developed Veppanu (vepdegestrant), the first FDA-approved PROTAC therapy, for adults with ER-positive/HER2-negative, ESR1-mutated advanced or metastatic breast cancer whose disease has progressed after at least one line of endocrine therapy.
Arvinas is currently progressing multiple investigational drugs through clinical development programs, including ARV-393, targeting BCL6 for relapsed/refractory non-Hodgkin Lymphoma; ARV-102, targeting LRRK2 for neurodegenerative disorders; ARV-027, targeting the polyglutamine-expanded androgen receptor, or polyQ-AR, in skeletal muscle for the treatment of Spinal-Bulbar Muscular Atrophy, also known as Kennedy's disease; and ARV-806, targeting KRAS G12D for mutated cancers, including pancreatic and colorectal cancers.
Nurix Therapeutics NRIX, a clinical-stage biopharmaceutical company, is also developing targeted protein degradation therapies for cancer and autoimmune diseases. NRIX's clinical pipeline includes bexobrutideg, a BTK degrader being co-developed with Roche for oncology and autoimmune indications; NX-1607, a CBL-B inhibitor for cancer; and an IRAK4 degrader partnered with Gilead for autoimmune diseases.
Nurix is also advancing a portfolio of preclinical protein degraders and degrader-antibody conjugates, supported by strategic collaborations with Roche, Gilead, Sanofi and Pfizer.

Shares of the company have gained 52.5% year to date compared with the industry’s growth of 9.7%.
